Malignant melanoma is currently the leading cause of death from cutaneous malignancies, and it is estimated that one out of 61 persons in United States will develop malignant melanoma in his or her lifetime. The incidence of melanoma has increased five-fold during the past three decades, and is rising faster than any other cancer in men and second only to lung cancer in women. Metastasized melanoma is extremely aggressive, often fatal, and refractory to current therapies. It has a very poor prognosis: the median survival rate is 6 months, and the 5-year survival rate is less than 5-15%. Therefore, there is pressing need to seek powerful chemotherapeutic agent to cure malignant melanoma.
My lab has been conducting several lines of research aiming to advance the understanding of melanoma biology and therapeutics. (1) My lab is currently studying the effects and working mechanisms of supplementary antioxidants like Vitamin C, β-carotene and co-enzyme Q10 on the growth and metastasis of human malignant melanoma and their effect on the therapeutic efficacy of FDA approved anti-melanoma BRAF and MEK inhibitors. (2) Also, my research group has been working on developing cocktail therapy regimen combining BRAF inhibitors with heat shock protein inhibitors and autophagy inhibitors to overcome the drug resistance in human malignant melanoma cells. (3) In addition, we also investigate the biological functions of microRNAs (miR-15a, miR-26a, let-7a, etc.) in growth, motility, invasiveness and apoptosis of melanoma cells, aiming to develop microRNA-based therapy against melanoma. We are currently developing a novel CRISP-Cas9 system for increasing the expressions of tumor suppressor microRNAs. (4) We are currently defining the differential activation patterns of p53 network in melanoma cell with different genetic context, with the goal of identifying the therapy targets for restoring the deregulated p53 network in cancers. (5) My lab screens and examines the pharmacological effects of a number of standard preparation of plant extracts and individual constituents of the extract on malignant melanoma. We elucidate the anti-melanoma mechanisms by examining the differential gene expression profiles of melanoma cells in response to phytochemicals. We also identify the genes and signaling pathways that phytochemicals target to exert the anti-melanoma effects by gene knockout through RNA interference.
In addition, our group has other three ongoing projects that my graduate students are currently working on: (1) Interactive effects between dam (methylation of GATC sites) deficiency mutation and the mutations of mutD, mutH, mutL, or mutS (genes required for methyl-directed mismatch repair). (2) Effect of mercuric compound at sublethal concentrations on the survivability of bacterial cells challenged by genotoxic agents (3) Characterizing a novel antimicrobial and anti-tumor agent produced by a strain of Brevibacillus laterosporus, a spore-former isolated from Rocky Mountain soil sample.
Due to my background in both molecular genetics and computer science, I am also interested in giving solutions to the mathematical or computing algorithm problem using bacteria cells, taking advantage of directionality of genes. My lab is working on designing innovative genetic circuits to facilitate topological search and solve Hamiltonian path problem.
